Pharmacokinetics and Pharmacodynamic of Multiple Intramuscular Dosage of Diminazene in infected goats

ABSTRAK

Telah dilakukan investigasi farmakokinetik dan farmakodinami diminazen terhadap 5 ekor kambing penderita Tripanosoma evansi isolat bangkalan dan 5 ekor kambing sehat. Pada uji klinik dosis yang digunakan adalah 10,22 mg.kg-1 berat badan (bb) sebagai dosis muatan dan empat kali tambahan dosis penjagaan (7 mg.kg-1 bb) tiap interval 31 menit. Tujuan penelitian ini adalah untuk mendapatkan data hubungan antara parameter kinetik dengan respon tripanosidal pada pemberian dosis berganda secara intra muskuler. Kromatografi cair kinerja tinggi fase terbalik, digunakan untuk analisis obat dalam plasma darah. Konsep superposisi dan ulas darah tebal digunakan untuk analisis parameter kinetik dan pengamatan eliminasi parasit. Card agglutinasi tes digunakan untuk penilaian keberadaan antigen Tryanosoma 45 hari pasca pengobatan.Paparan parasit terhadap lamanya konsentrasi terapi mobat pada rentang waktu rerata keberadaan obat antara 5,20 sampai 14,66 hari dihasilkan dari pemberian dosis berganda. Data tersebut menunjukkan bahwa Tripanosoma evansi isolat Bangkalan umumnya relatif sensitif terhadap diminazen dosis berganda (p<0,05).

ABSTRACT

The pharmacokinetics and the pharmacodynamic of diminazene had been investigated in 5 infected goats of Trypanosoma evansi bangkalan isolates and 5 of healthy goats. The 10.22 mg.kg-1 bw as loading dose and fourth times addition of maintenance dose (7 mg.kg-1 bw) each 31 minutes intervals was using for clinical trials. The objective of this research was to obtained realtionships data of kinetics parameter vs., trypanocide response at multiple dosng intramuscularly administrations. The high performance liquid chromatography reverse-phase was used for analysis drug concentration in blood plasma. The superimpose concepts and thick blood film method were used for analysis kinetic parameterand assessment of parasites elimination. The card agglutination test was used for assessment of available trypanosomes antigen 45 days post treatment.The expore of parasites to length of therapeutics drug concentration at ranging Mean Residance Time of 5.20 to 14.66 days was resulting multiple dosing. These data indicate that population of Trypanosoma evansi Bangkalan isolates, was generally sensitive to multiple dosing of diminazene (p<0.05).
